AbISCO®-300
-- a cost effective preparation for use in larger animals
A selection of purified fractions of quillaja saponins formulated with a mixture of cholesterol (ovine wool) and phosphatidyl choline (egg). The product is sterilized by filtration through 0.22 µm sterile filter. The material is tested for endotoxin. If required, the preparation can be sterile filtered again without significant loss of material.
Note: AbISCO®-300 is a research adjuvant -- not for human use.
Easy to use with any antigen. A solution of AbISCO®-300 is non-viscous; no adsorption of antigen or emulsification process is necessary. Just mix an appropriate amount of AbISCO®-300 with the antigen in PBS or saline. AbISCO®-300 a cost effective preparation suitable for a wide range of larger animal species e.g., sheep, goat and cow.
The optimum dose of AbISCO®-300 depends on the antigen, the animal species and the mode of administration. As a general rule, small hydrophilic antigens (poor antigens) require more adjuvant than larger antigens, particulate antigens such as whole killed bacteria or viruses, protein micelles, or antigens that are prone to aggregation.
Stable more than 24 months at +2 to +8°C. To ensure stability of the saponins, do not store adjuvant/antigen solutions at pH exceeding pH 7.4.
Toxicity of saponin preparations has been a controversial issue surrounding the development of saponin-based human and veterinary vaccines. Different toxicity results have been reported, most likely due to the complication of different grades of quillaja saponins and difficulties in many laboratories in assessing the actual amount of saponin in, e.g., an ISCOM preparation. AbISCO® adjuvants are made from selected saponins and the amount is carefully analysed. This makes AbISCO® adjuvants safe for use via subcutaneous or intramuscular administration. Formulation into an ISCOM particle reduces the inherent lytic activity of the quillaja saponins to minimum.
Note: Intravenous, intraperitoneal or foot-pad routes of administration are not recomended.
